We investigated the effect of polysaccharide rich Pleurotus pulmonarius fruit body extracts on progression of chemically induced hepatocellular carcinoma in CBA mice. This study was designed to evaluate the influence of manure and mineral fertilization on yield, Kjeldahl nitrogen (as protein equivalents), Vitamin C and nitrate accumulation in Amaranthus hypochondriacus. The vegetables were produced in field trials, set up in the University of Nairobi, Kenya, during the long rains period between March and May in 2007 and 2008 using diammonium phosphate (DAP; 20, 40 and 60 kg N ha-1) and manure; 40 kg N kg ha-1,) and the yield, Kjeldahl nitrogen (K-N; a proxy for protein content), vitamin C and nitrate levels were measured. The manure treatment produced little increase in productivity though it increased K-N levels. The DAP treatments produced increases in yield, but these saturated at 40 kg N ha -1. The K-N content of the leaves was high, ranging from 2.5 % to just over 4 %, and, broadly, increased with fertiliser application though the variation between the treatments was lower than that for yields. Nitrate levels, however, also increased with DAP fertilisation, resulting in an increase in the nitrate:K-N at high DAP applications, especially in younger plants. There were no clear correlations between ascorbic acid content and K-N. Nonetheless, the plants grown under different fertilisation regimes can be placed into three broad groups; those characterised with low to moderate K-N contents and low to high ascorbic acid contents, moderate to high K-N and low to high ascorbic acid content, and lastly high K-N and low ascorbic contents. Therefore, the growth environment of amaranth, can have a major impact not only on the productivity of the crop but its nutritional value. In this respect the mineral nutrient supply to the crop is of particular importance.

This study was undertaken to characterize the alpha subgroup of the proteobacteria causing the huanglongbing (HLB) disease of citrus from three different ecological zones of Kenya namely the Lower highlands (LH2, LH3, 1800-1900 m above sea level); Upper midlands (UM3, UM4, 1390-1475m), Lower midlands (LM5, LM4, LM3 of 1290-1340-1390m), by isolation and sequencing DNA encoding the L10 and L12 ribosomal proteins and the intergenic region. A 7I6-basepair DNA fragment was amplified and sequenced and consisted of 536 basepairs of DNA encoding the L10 protein, 44 basepairs of DNA intergenic region and 136 basepairs of DNA that partially encodes the L12 protein. Sequences of rpL10/L12 protein genes from Kenyan strains were 98% and 81% similar to the South African ‘Candidatus Liberibacter africanus strain Nelspruit' and the Asian ‘Candidatus Liberibacter asiaticus' strains, respectively. The intergenic rDNA sequence of Kenyan strain from UM and LM showed 84% similarity with ‘Candidatus L. africanus strain Nelspruit' and 50% similarity with ‘Candidatus L. asiaticus' strain. However, the LH strain had an 11- basepairs deletion, while the LM4 had a 5- basepair deletion in the intergenic region compared to ‘Candidatus L. africanus strain Nelspruit'. The L10 amino acid sequence was 100% homologous among HLB bacteria obtained from the agro-ecological zones in Kenya and the L10 protein sequence was also homologus to ‘Candidatus L. africanus strain Nelspruit'. Nevertheless, the L10 amino acid sequence of ‘Candidatus L. asiaticus' and the ‘Candidatus L. africanus subsp. capensis' differed from the Kenyan strains by 18.36% and 11.82%, respectively. Phylogenetic analysis of both the L10/L12 rDNA sequences and the L10 amino acid sequences clustered the Kenyan strains of the ‘Candidatus Liberibacter' species with members of alpha subdivision of proteobacteria.

Potato (Solanum tuberosum L.) genotypes (Tigoni, Asante and Dutch Robyjn) were evaluated for quality under three light intensities, 612.2 kW (diffused), 1376 kW (direct) and 8 kW (dark) for 12 weeks. Tuber stored in dark conditions had a lower weight loss than tubers stored in direct light conditions. Dutch Robyjn lost the lowest weight (4.49%) while Asante lost the highest (13.90%) mean weight. Tigoni in the dark had the highest number (9.25) of sprouts. Tubers in the dark developed long (46.25mm) etiolated sprouts as opposed to the short (10.50mm) firm sprouts observed in tubers subjected to the diffused light. High sprout vigor score (2.42) was observed in Asante while Dutch Robyjn had the lowest (1.00) score. Nevertheless, all the tubers in the three light intensities had 100% sprouted tubers after the 12 weeks of storage. Potato tuber moth incidence was higher in percentage in the direct and diffused light than in the dark storage. No incidence of rotting was reported for the 12 weeks of storage. It is therefore suggested that farmers should adopt storage of potato seed tubers in diffused light (612.2 – 1000 kW) store to allow formation of short firm sprouts and reduce weight loss.

Menza, Mwalimu K; Shibairo SNRDP; KJGPI; O; N. Effects of pit, dark and cold pre-storage treatments and their duration on dormancy breaking and sprouting of seed potato tubers (Solanum tuberosum L.).; 2008. Abstract

Potato is the second most important food crop after maize in Kenya. In the traditional areas of potato production with a bimodal rainfall pattern in the country, poor sprouting, due to seed tuber dormancy, is a major drawback. There is little time between growing seasons to permit adequate sprouting of the seed tubers. Therefore, effects of pit, dark and cold pre-storage treatments and their duration on dormancy breaking and sprouting of seed potato tubers of variety Asante were determined. Tubers were evaluated for sprouting, number of sprouts per tuber and sprout vigor for 12 weeks. Pit and dark pre-storage treatments resulted in significantly higher sprouting, number of sprouts per tuber and vigor scores than cold pre-storage treatment and the control (diffused light storage). 100% sprouting of seed potato tubers was attained under pit storage by the fourth week for all pre-storage treatment durations while dormancy ended after 6 weeks of storage in the control. Sprouting was suppressed during cold pre-storage treatment. In pit and dark pre-storage treatments, vigor scores increased with increasing duration of pre-storage treatment while in the cold, vigor scores were reduced with longer pre-storage treatment duration. Pit and dark pre-storage treatments for short durations of up to one week respectively followed by two weeks of diffused light storage are recommended to break dormancy and promote sprouting of good quality seed potato tubers of Asante variety.

Lung’aho CG, Chemining’wa S, Shibairo, Hutchinson M. "Reaction of Potato Cultivars to Natural Infestation of Late Blight Caused by Phytophthora infestans in Kenya.". 2008. Abstract

The late blight resistance and yield characteristics of 32 potato genotypes were studied at KARI-Tigoni between 2006 and 2007 under natural late blight inoculums pressure. Foliar late blight resistance was assessed on the basis of area under disease progress curve (AUDPC) whereas the relative area under disease progress curve (RAUDPC) was used to classify the genotypes into five late blight resistance groupings (resistant, moderately resistant, intermediate, moderately susceptible and susceptible). Tuber yields for individual cultivars ranged from 5.87 to 60.70 t/ha. The correlation between foliar late blight and total tuber blight was weak but that between foliar late blight and total tuber yield was negative and significant. On the basis of RAUDPC, it was possible to classify 26 of the 32 cultivars into distinct resistance classes. Cultivars Kenya Karibu, Tana Kimande, Kihoro, Kenya Sifa and Kenya Karibu (white flowers) were rated as resistant. The most susceptible cultivars were Kerr’s Pink, Ngure, Desiree, Romano and Pimpenel. Any of the resistant or susceptible varieties can be used as checks in evaluating germplasm for late blight tolerance.

The effect of potassium (K) nutrition on the shelf life of carrots was studied using a hydroponics system involving rockwool slabs as support. Carrots were grown for 192 days under greenhouse conditions and supplied with 0, 0.1, 1.0, 10, and 15 mm of K. Increase in K concentration in the nutrient medium decreased postharvest weight loss. Carrot weight and tissue K content increased and water potential, osmotic potential, and relative solute leakage decreased with increasing K concentration in the nutrient feed. Differences in postharvest weight loss were mainly associated to root weight and relative solute leakage. Root weight correlated negatively and relative solute leakage correlated positively to water loss. Water and osmotic potential also correlated to water loss, but not as strongly as root weight and relative solute leakage. These results suggest that K nutrition influences postharvest weight loss by influencing carrot size and membrane integrity. Effects on cell water and osmotic potential are also important in this regard but to a lesser extent.

Studies were carried out to understand the effects of moisture loss on water potential and root deterioration in carrot (Daucus carota L. `Eagle') roots during short-term storage. The roots were stored at various temperatures and relative humidities (RH) to provide 0.7 (low), 3 (medium), and 9 mbars (high) of water vapor pressure deficit (WVPD). Carrots at high WVPD lost the most weight, followed by those at medium and lowest WVPD. Water potential and osmotic potential of the carrot tissue at high WVPD did not change significantly up to 6 days, but decreased thereafter. There was no change in water potential and osmotic potential for carrots at medium and low WVPD. A significant quadratic relationship (P = 0.05, r = –0.764) between water potential and carrot root weight loss was observed. Relative electrolyte leakage increased over time in carrots at the high WVPD. At medium WVPD, relative electrolyte leakage did not change up to 6 days, but increased significantly thereafter. Carrots at the low WVPD did not change in relative electrolyte leakage. Relative electrolyte leakage and weight loss correlated positively (P = 0.05, r = 0.789). The results suggest that water stress during short-term storage causes tissue deterioration that may further increase rate of moisture loss and hence reduce the shelf life of carrots.

Two sets of incubator experiments to determine the effects of temperature on germination of three pigeonpea genotype seeds were carried out in the Crop Science Laboratory of the Faculty of Agriculture, University of Nairobi. The pigeonpea genotypes were NPP 670, Katheka and Kioko. In the first set of experiments, the pigeonpea seeds were germinated at 15, 20,25,30,35 and 40°C in darkness. In the second set of experiments the seeds were germinated at different 12 hour day and 12 hour night temperature combinations of 15/15, 20/15, 25/15, 30/15, 20/20°C, 25/20, 30/20 and 35/20°C for day and night, respectively. A broad temperature range (20-35°C and 20/15°C to 30/20°C day/night) maximum 13-day germination was obtained for all genotypes. Overall genotype NPP 670 had the highest per cent germination while Kioko had the lowest. However, Kioko had the highest per cent germination at 15°e. Initial germination was delayed at 15°C and 40°C for all genotypes. Time to 50 per cent germination was only Significantly (P = 0.05) increased at 15°C. Maximum per cent germination was observed at 25°C. At 40°C, genotypes Kioko and Katheka seeds imbibed water at a faster rate and exuded brownish substances that started smelling after 36 hours. A bacterial ooze from the seeds was observed for genotype Ka theka at 30°C, 20/15 and 25/ 20°e. NPP 670 showed low amounts of the bacterial ooze at all temperatures. The bacteria fluoresced under ultraviolet light suggesting that they belonged to the Pseudomonadaceae family. The results showed that per cent germination of pigeonpea seeds is decreased by both low and high temperatures. Other factors which include presence of seed borne micro-organisms may also affect the germination of seeds
